A biosensor was crafted by incorporating LC3B flanked within a pH-resistant donor-acceptor FRET pair, Aquamarine-tdLanYFP. We found the biosensor to have a dual readout, as evidenced by our analysis. The priming of LC3B by ATG4B is shown through FRET, enabling the detailed examination of the spatial differences in priming activity through the resolution of the FRET image. Secondly, an evaluation of autophagy activation is based on the count of Aquamarine-LC3B puncta. A decrease in ATG4B led to the accumulation of unprimed LC3B, and priming of the biosensor was not observed in ATG4B knockout cells.
